摘  要:目的应用代谢组学技术分析肺癌患者和正常人群血清的代谢物质。方法运用高效液相-四级杆-飞行时间质谱(LC—Q—TOF/MS)联用技术对30例肺癌患者与30名正常人血清进行研究。采用正交最小二乘分析法(OPLS)进行建模,运用两样本t检验寻求两组间差异性代谢物。结果多变量统计结果显示肺癌患者和正常对照人群的代谢谱有明显差异,根据t检验结果寻找到血清中相关的差异代谢物36种。结论建立了利用LC—Q—TOF/MS技术进行肺癌血清代谢组学研究的方法。Objective To explore the differences in serum metabolism between patients with lung cancer and healthy people using the technology of metabolomies. Methods Serum samples from 30 lung cancer patients and 30 healthy people were analyzed by high performance liquid chromatography-quadrupole time-of-flight mass spectrometry (HPLC-Q-TOF/MS). Orthogonal to partial least squares discriminant a nalysis was performed for modeling. Two sample t-test was used to identify differences in metabolite concentrations. Results Multivariate statistical analysis yielded a significant distinction in the metabolic profiles between lung cancer patients and healthy people. The t-test results indicated a total of 36 metabolites in the serum with statistically significant differenees. Conclusion The method of lung cancer serum metabolomics study has been constructed using the technology of HPLC-Q-TOF/MS.
