摘 要:目的 研究c met SF/HGF基因表达对肝癌细胞转移能力的影响。方法 采用RT PCR及WesternBlot法检测肝癌细胞株MHCC 1、HepG 2、Hep3B、SMMC772 1的met SF/HGF的表达 ,继而用c met多克隆抗体阻断met SF/HGF的信号传导 ,观察细胞各项能力的改变。结果 转移能力高的细胞株 (MHCC 1)c met的表达量远高于转移能力低的细胞株 ,并伴有配体SF/HGF的自分泌 ;MHCC 1上清液能刺激SMMC772 1细胞增殖并使其运动能力增强 ;该作用可被抗c met抗体特异性阻断 ,当被阻断后 ,细胞生长及运动能力下降。结论 met SF/HGF基因表达在肝癌转移中起重要的作用 ,阻断c metObjective To clarify the function of c met and its ligand HGF expression in hepatocellular carcinoma.Methods RT PCR and Western Blot were used to examine the expression of c met and SF/HGF in hepatocellular carcinoma cell line MHCC 1,HepG2,Hep3B and SMMC7721.SMMC7721 was also stimulated by the conditioned medium of MHCC 1,and the stimulation was inhibited by c met polyclonal antibody to observe cell mobility and proliferation.Results Cell mobility of hepatocellular carcinoma was relative to its met SF/HGF expression.The expression in cell line with higher potential of metastasis(MHCC 1) was much stronger than in those with lower potential of metastasis and was with SF/HGF autocrine.An increase of proliferation and mobility happened in SMMC7721 after it was stimulated by conditioned medium of MHCC 1.Such functions could be specifically blocked by anti c met polyclonal antibody.Conclusion c met SF/HGF autocrine and paracrine play an important role in the metastasis potential of hepatocellular carcinoma.Inhibition of c met SF/HGF signal transduction system may reduce the metastasis of hepatocellular carcinoma.
