TNF-α、M-CSF及IL-1在骨巨细胞瘤中的表达及其意义

摘　　要：目的　探讨细胞因子与骨巨细胞瘤局部溶骨的关系。方法　采用ELISA法、Westernblot分析和免疫组化双染检测了 10例骨巨细胞瘤、5例骨肉瘤和 5例正常人血清中TNF α含量和M CSF、IL 1的表达。结果　骨巨细胞瘤组织TNF α含量和M CSF表达率明显高于骨肉瘤和正常人。TNF α由骨巨细胞的部分单核基质细胞和多核巨细胞分泌 ;而M CSF及IL 1由部分单核基质细胞分泌 ,多核巨细胞则不表达。Objective To study the relationship of the expression of TNF α,M CSF and IL 1 and local osteolysis of giant cell tumor of bone.Methods Expressions of TNF α,M CSF and IL 1 were detected by ELISA,Western blot analysis,immunohistochemical staining method in 10 cases of giant cell tumor of bone,5 cases of osteosarcoma and 5 cases of normal serum.Results There was higher level of TNF α and higher expression rate of M CSF in giant cell tumor compared with in osteosarcoma and normal serum.TNF α was expressed in part of mononuclear stromal cells and in multinucleated giant cell,whereas M CSF and IL 1 was only expressed in part of mononuclear stromal cells.Conclusion The cytokines secreted by various cells of giant cell tumor of bone might be involved in local osteolysis of the tumor.
