摘 要:目的采用柱前衍生RP—HPLC法对黄芩中游离及总氨基酸进行测定。方法以异硫氰酸苯酯为柱前衍生化试剂;色谱柱为Ultimate Amino Acid氨基酸分析专用柱(4.6mm×250mm,5μm);流动相:A为乙腈-水(80:20,WV),B为三水合醋酸钠缓冲溶液(pH6.5)-乙腈(93:7,V/V),梯度洗脱;检测波长为254nm。结果21种氨基酸在0.0025~2.5000mmol/L内呈良好的线性关系(R^2〉0.9990)。21种游离氨基酸平均回收率在93.72%~105.03%之间,RSD在0.81%~3.15%之间(n=6);总氨基酸平均回收率在95.22%~102.04%之间,RSD在0.47%~2.44%之间(n=6)。结论柱前衍生RP-HPLC法灵敏、准确,具有良好的重复性和稳定性,适合黄芩中氨基酸含量测定。Objective To determine the content amino acid in Radix ScuteUariae by pre-column derivatization RP-HPLC. Methods The samples were derived with phenylisothiocyanate (PITC). The column was Ultimate Amino Acid of specific column (4.6 mm × 250 mm, 5μm) ; Mobile phase: A is mixture of acetonitrile-water ( 80 : 20, V/V) , B is a buffer solution of sodium acetate trihydrate ( pH 6.5 ) -acetonitrile (93 : 7, V/V) , eluting with a gradient. UV wavelength was set at 254 nm. Results The linear range of 21 amino acids was 0. 002 5 - 2. 500 0 mmol/L ( R2 〉 0. 999 0) ; the average recovery rates of 21 free amino were ranged from 93.72% to 105.03% ( RSD = 0.81% - 3.15%, n = 6), the average recovery of rates of 21 total amino acids was ranged from 95.22% to 102.04% (RSD =0.47% -2.44%, n = 6 ). Conclusion This method is flexible, accurate and repeatable and stable for the determination of amino acids in Radix Scutellariae.
