摘 要:目的探讨AKT/mTOR信号转导途径中AKT和PTEN基因在非小细胞肺癌(NSCLC)组织中的表达和临床意义。方法外科手术中获取72例NSCLC组织及30例癌旁组织,逆转录聚合酶链反应C(RT-PCR)技术检测NSCLC组织及癌旁组织中AKT和PTEN基因的表达水平。结果 AKT在NSCLC组织的表达量(0.24±0.15)显著高于癌旁组(0.11±0.08),PTEN在NSCLC组织中表达量(0.18±0.27)显著低于癌旁组(0.54±0.27),P均﹤0.05。AKT和PTEN与患者的性别、年龄、病理类型、淋巴结转移情况无关,与肿瘤直径有关。结论 AKT通路的激活和PTEN表达缺失在NSCLC发生、发展中起到一定作用。Objective To detect the mRNA expression levels of AKT and PTEN gene, which are the key genes of AKT/mTOR signaling pathway in human non-small cell lung cancer (NSCLC) tissue. Methods NSCLC tissue specimens were obtained from 72 patients. Adjacent-tumor NSCLC tissues from 30 patients were used as controls. RT-PCR was used to detect the AKT and PTEN gene expression levels. Results The levels of AKT gene expression in NSCLC tissue were significantly higher than those in adjacent-tumor lung tissue [(0.24±0.15) vs. (0.11±0.08), P 〈 0.05]. The levels of PTEN gene expression in NSCLC tissue were significantly lower than those in adjacent-tumor lung tissue [(0.18±0.27) vs. (0.54± 0.27), P 〈 0.05]. There were no significant relationship between AKT, PTEN gene expression levels and patients age, gender, pathological type,differentiation, lymph node metastasis, except tumor size. Conclusion The absence or decreased expression of PTEN and the activated AKT may play an important role in the carcinogenesis and metastasis of NSCLC.
