间皮素mRNA及蛋白在上皮性卵巢癌中的意义研究

摘　　要：目的研究mesothelin(间皮素,MESO)在上皮性卵巢癌中的表达及其与卵巢癌发生、发展之间的关系。方法采用半定量RT-PCR技术及免疫组化方法分别检测上皮性卵巢癌,卵巢肿瘤和正常卵巢组织中MESO mRNA及其蛋白的表达情况,并分析其表达在不同临床分期和病理分级之间的关系。结果上皮性卵巢癌中MESO mRNA(1.4005±0.4646)及蛋白(2.7857±2.2712)表达水平明显高于良性卵巢肿瘤(0.6463±0.2419,1.2500±1.6125)和正常卵巢组织(0.6439±0.2729,0.9167±1.2401),差异有显著性(P<0.05);MESO mRNA及蛋白在浆液性卵巢癌(1.5255±0.4151,3.3036±2.6141)和子宫内膜样癌(1.5250±0.5419,3.0000±2.3094)中的表达高于其在粘液样癌(1.0675±0.3149,1.0566±1.9242)中的表达,差异有统计学意义(P<0.05);MESO在临床Ⅲ、Ⅳ期卵巢癌中的表达显著高于其在Ⅰ、Ⅱ期卵巢癌中的表达,差异有统计学意义(P<0.05);MESO的表达水平与病理分级、病理类型有关,差异有显著性(P<0.05);MESO的表达水平与患者有无腹水有关(P<0.05),而与患者的年龄、血清CA125水平无关(P>0.05)。结论上皮性卵巢癌MESO mRNA及蛋白的表达增高,MESO可能参与卵巢癌的细胞识别、黏附、转移,并促进其腹腔播散与种植。Objective：To investigate mesothelin（MESO）mRNA and protein expressions in the epithelial ovarian carcinoma and their effects in the tumorigenesis and tumor progression of ovarian carcinomas. Metheods：The expressions of MESO mRNA and protein were examined by semi-quantitative reverse transcription polymerase chain reaction（RT-PCR）and immunohistochemistry respectively in the epithelial ovarian carcinomas,benign ovarian tumor and normal ovarian tissues.The clinical characteristics were compared and clinical significance was analyzed.Result：The mRNA（1.4005±0.4646）and protein（2.7857±2.2712）expressions of MESO in epithelial ovarian carcinoma were significantly higher than in benign ovarian tumor（0.6463±0.2419,1.2500±1.6125）and normal ovarian tissues（0.6439±0.2729,0.9167±1.2401）（P0.05）,and also significantly higher in serous cystadenocarcinoma（1.5255±0.4151,3.3036±2.6141）and endometrioid carcinoma（1.5250±0.5419,3.0000±2.3094）than that in mucinous cystadenocarcinoma（1.0675±0.3149,1.0566±1.9242）（P0.05）;The expressions of MESO mRNA and protein in stages Ⅲ and Ⅳ carcinoma was significantly higher than that in stagesⅠand Ⅱcarcinoma（P0.05）;The expression of MESO was related to grading,histologic subtype and the amount of ascites（P0.05）,but not to the patients age or serum CA125（P0.05）.Conclusion：Our findings indicate that MESO mRNA and protein are increased expression in epithelial ovarian carcinoma.Mesothelin may play a role in cell recognition,adhesion,metastasis and facilitate the dissemination and implantation of tumors in the peritoneal cavity.
