摘 要:目的探讨P-选择素对小鼠黑色素瘤肺转移的影响。方法将体外培养的B16黑色素细胞株通过小鼠的尾静脉注射到小鼠背景为C57/BL(C57)的P-选择素敲除的小鼠和C57小鼠的体内,建立转移瘤瘤模型,第21天断颈处死小鼠,计数转移到肺表面的结节的数目,HE染色进一步确定转移结节。结果 P-选择素敲除小鼠肺表面转移的肿瘤结节的数目明显比C57小鼠少,两者比较差异有统计学意义(P﹤0.01)。HE染色证实小鼠肺组织中有B16细胞的肺转移灶。结论 P-选择素缺失可能参与抑制了黑色素瘤的肺转移。Objective To study the effect of P-selectin on pulmonary metastasis of melanoma in mice. Methods B16 cells were intravenously injected into P-selectin knockout and C57/BL( C57 )mice. After three weeks, the mice were sacrificed by cervical dislocation. Lung metastasis was observed by gross anatomy. HE staining of the metastatic tumor tissue sections was performed to confirm homology with B16 cells. The number of metastatic foci was counted. Results P-seleetin knockout mice developed fewer metastatic foci on the lung surface than C57 mice(P 〈 0. 01 ). HE staining of the metastatic tumors confirmed their homology with B16 cells. Condusion P-seleetin loss may be involved in the inhibition of melanoma metastasis to the lung.
