不同储藏条件下鳜鱼样品中孔雀石绿及隐色孔雀石绿的降解规律研究

摘　　要：鳜鱼稀少、名贵、难养，是孔雀石绿（MG）及其主要代谢物隐色孔雀石绿（LMG）残留重灾区。为确定鳜鱼样品中用于开展能力验证药物的适宜储存条件和运输温度条件，最大限度消除外因对MG及LMG检测结果的影响，保证结果的可靠性，以鳜鱼肌肉为研究对象，开展了MG及LMG残留降解规律研究。实验分析了MG和LMG在3种浓度（5.0、10.0、50.0μg/kg）和3种贮藏条件（室温20～25℃，冷藏0～4℃和冷冻-18℃）下随时间的降解规律。实验结果表明，不同浓度的MG和LMG均有不同程度的降解，但因贮藏条件的不同，降解速率各不相同。在各种贮藏条件和不同浓度加标情况下，鳜鱼样品中LMG代谢消除速率均显著慢于MG；且因LMG的毒性远大于MG，建议将LMG作为残留检测的主要标志物。受酶活性高低的影响，贮藏条件对MG和LMG代谢的影响较大，降解速率为常温＞冷藏＞冷冻。如果样品能尽快进行检测，即可在常温条件下进行短期保存；如样品不能尽快安排检测，则须冷冻保存，尽量保持样品的原有属性。Mandarin fish is a kind of rare fish which is famous for the delicious taste and the difficulty of feeding it. It is the worst-hit area of the malachite green （MG） and leuco malachite green （LMG）, the primary metabolite of MG. In order to filter out the appropriate storage and transportation temperature conditions for drug verification in mandarin fish, minimize the impacts of external factors on the test results and ensure the fairness of the outcome, mandarin fish muscle was used as the study object to analyze the degradation of MG and LMG over time in three different concentrations （5.0, 10.0, 50 μg/kg） and under 3 different storage conditions （room temperature 20～25℃, cold storage 0～4℃ and freezing -18℃）. The results showed that MG and LMG of different concentrations degraded with various degrees, however, the degradation rate varied due to different storage conditions. In different storage conditions and standard addition concentrations, the metabolic elimination rate of LMG is significantly slower than that of MG; moreover, the toxicity of LMG is far larger than ML, so LMG was suggested to be employed as the main marker for the residue detection. Due to the impact of enzyme activity, storage conditions had a great influence on the metabolism of MG and LMG. The result was showed with the degradation rate of room temperature condition 〉 refrigerated condition 〉 freezing condition. Malachite green samples could be kept at room temperature for a short time, if the detection was to be conducted soon. If samples could not be detected in a short time, they should be preserved by freezing method to remain their original properties.
