摘 要:目的探讨抗性淀粉(RS)对饮食诱导肥胖(DIO)大鼠解偶联蛋白2(UCP2)基因表达水平的影响,为开发RS相关保健食品提供科学依据。方法选择SD大鼠60只,体重(100±10)g,雌雄各半,随机分为对照组和高脂组,分别用普通饲料和高脂饲料喂养7周。根据体重筛选出DIO大鼠,再随机分成4组,分别用高脂饲料、含5%、10%及15%RS高脂饲料喂养5周后,检测各组实验动物体重、脂体比及白色脂肪组织中UCP2mRNA的表达水平。结果 RS添加组DIO大鼠体重和脂体比呈现下降趋势,UCP2mRNA表达升高,与高脂饲料喂养的肥胖对照组比较差异均有统计学意义(P<0.05),且RS添加量与体重呈负相关(r=-0.862,P=0.000)。结论 RS可增强UCP2基因表达,减少能量储存,有一定的减肥效果。Objective To examine the effect of resistant starch (RS) on the expression of uncoupling protein 2 (UCP2) in diet-induced obese (DIO) rats in order to provide scientific evidence for developing RS-related health foods. Methods Sixty SD rats i-weight(100 ± 10)g] with even distribution of males and females were divided into control group and high-fat group at random. The rats in the control group were fed on a normal food for 7 weeks and those in the high-fat group on a high-fat food for the same period of time. DIO rats were selected according to the weight and then divided randomly into 4 groups,in which the rats were fed on the higH-fat food,and the high fat food containing 5o//00 RS,10~ RS,or 15~ RS,respectively for 5 weeks. The weight,the fat-to-weight ratio, and the expression level of UCP2 mRNA in white adipose tissues were detected in each group. Results The DIO rats fed on RS-contained foods were found to have decreased weights and fat-to-weight ratios and in- creased expression levels of UCP2 mRNA. There was significant difference in these variables between rats fed on high fat food containing RS and those on high fat food only (P〈0.05). The dose of the added RS was found to be inversely correlated with the weights of rat (r=-0. 862,P=0. 000). Conclusion RS has a certain effect on weight loss by increasing the expression of UCP2 gene and reducing energy storage.
