摘  要:合成了一种以壳聚糖为载体 ,以正丁基为配基 ,并以戊二醛交联提高其机械强度的新型疏水色谱填料—丁基交联壳聚糖 ,并对其合成条件的控制进行了研究。结果表明 :交联剂用量、反应物比例以及反应物浓度决定了合成填料的疏水性能。为保证良好的操作性、实用性及较佳的分离效果 ,比较合适的合成条件是 :戊二醛浓度 0 .1 % (V/V ) ,正丁醛浓度 0 .1 3 %~ 0 .2 6% (V/V ) ,壳聚糖浓度 2 g/L~ 1 0g/L。不同条件下合成的色谱填料对枯草杆菌α-淀粉酶粗品的分离提纯效果明显不同 ,进一步说明合成条件对色谱填料性能的影响。A novel hydrophobic interaction chromatographic(HIC) packing material has been synthesized by inducing butyl group to chitosan. In order to harden the HIC packing material, chitosan was cross linked by gluteraldehyde firstly. The butyl group was induced by the Schiff reaction of butyraldehyde to the free amino group of chitosan. The synthesizing conditions, particularly the concentrations of butyraldehyde and chitosan were investigated. The results showed that the concentrations of reactants affect the hydrophobic properties of the packing material significantly. In view of practice, the recommended concentrations of three reactants are 0.1% gluteraldehyde (V/V), 0.13%-0.26% butyraldehyde (V/V) and 2 g/L-10 g/L chitosan. However, the exact concentrations are dependent on the purified protein because different proteins have different hydrophobicity. Two kinds of chitosan HIC packing materials synthesized under different concentrations of butyraldehyde were used to purify crude ɑ-amylase (from Bacillus Subtilis). The results further demonstrated that the purification efficiency of the HIC packing material is dependent on their synthesizing conditions.
