摘 要:目的:观察疏肝饮对内脏高敏感性大鼠结肠组织及DRG中SP和CGRP表达的影响。方法:96只wistar雄性大鼠随机分为正常组、模型组、疏肝饮大剂量组、疏肝饮中剂量组、疏肝饮小剂量组、西药组,采用慢性避水应激法造模10 d,造模第4 d起,各给药组大鼠于造模前半小时分别给予药物灌胃,连续7天,第10 d造模结束后,采用结直肠气囊扩张引起的腹壁撤回反射来评估内脏敏感性;取出结肠及DRG组织,采用western-blotting法分别检测SP和CGRP含量。结果:与正常组相比,模型组结肠及DRG中SP及CGRP的含量显著增加;与模型组比较,疏肝饮组大、中剂量组和得舒特组大鼠结肠及DRG中SP和CGRP含量均明显减少,差异有统计学意义。结论:疏肝饮通过调节结肠及DRG中SP和CGRP的表达,从而降低内脏高敏感性。Objective: To investigate the effects of Shugan Decoction on SP and CGRP in the colon and DRG of rats with visceral hypersensitivity. Methods: Ninety-six male Wistar rats were randomly divided into six groups: control group,model group,low dose Shugan Decoction group,middle dose Shugan Decoction group,high dose Shugan Decoction group and dicetel group. Rats were exposed to 1-h water avoidance stress( WAS) daily for 10 consecutive days. Shugan Decoction groups and dicetel group were given intragastric administration half an hour before WAS from the 4th day to the 10th day. The abdominal withdrawal reflex( AWR) to colorectal distension was assessed at the end of the 10-day period. Western-blotting method was applied to investigate the contents of SP and CGRP in the colon and DRG. Results: Compared with the normal group,SP and CGRP in the colon and DRG of model group were significantly increased. Compared with the model group,SP and CGRP in the colon and DRG of middle dose Shugan Decoction group,high dose Shugan Decoction group and dicetel group were significantly reduced. Conclusion: Shugan Decoction can reduce visceral hypersensitivity by regulating expression of SP and content of CGRP in the colon and DRG.
