摘 要:目的利用紫外分光光度法测定温肺化纤汤中总黄酮、总多酚、总皂苷和总多糖的含量。方法采用硝酸铝显色法测定温肺化纤汤中总黄酮的含量;采用Folin-ciocalteu法测定温肺化纤汤中总多酚的含量;采用香草醛-冰醋酸-高氯酸显色法测定温肺化纤汤中总皂苷的含量;采用硫酸苯酚比色法测定温肺化纤汤中总多糖的含量。结果芦丁在25∽200μg·ml-1范围内线性关系良好(r=0.999 0),测得温肺化纤汤中总黄酮含量为(7.37±0.76)mg·ml-1;没食子酸在10∽100μg·ml-1范围内线性关系良好(r=0.999 5),测得温肺化纤汤中总多酚含量为(11.83±0.12)mg·ml-1;薯蓣皂苷元在25∽200μg·ml-1范围内线性关系良好(r=0.999 7),测得温肺化纤汤中总皂苷含量为(20.75±2.43)mg·ml-1;葡萄糖在10∽100μg·ml-1范围内线性关系良好(r=0.999 0),测得温肺化纤汤中总多糖含量为(154.81±3.00)mg·ml-1。结论紫外分光光度法简捷、准确、灵敏度高,可以很好地测定温肺化纤汤中黄酮类、多酚类、皂苷类和多糖类化合物的含量,其结果为温肺化纤汤的新药研发提供了科学依据。Objective To determine the contents of major components in Wenfeihuaxian formula. Methods The contents of total saponin from Wenfeihuaxian formula were determined by vanillin - ice acetic acid - perchloric method. The contents of total sapo- nin from Wenfeihuaxian formula were determined by vanillin - ice acetic acid - perchloric method. The contents of total saponin from Wenfeihuaxian formula were determined by vanillin - ice acetic acid - perchloric method. The contents of total saponin from Wenfeihuaxian formula were determined by vanillin - ice acetic acid - perchloric method. Results The regression equation of rutin was A =0. 0026C -0. 00152 (r = 0. 999 0). The results showed that total flavonoids of Wenfeihuaxian formula was (7.37 ± 0.76 )mg·ml-1 The regression equation of gallic acid was A = 0.0095 C ± 0. 0248 ( r = 0.999 5 ). The results showed that total polyphenols of Wenfeihuaxian formula was ( 11.83 ± 0.12)mg·ml-1. The regression equation of diosgenin was A = 2. 2540C ± 0.0544 ( r = 0. 999 7). The results showed that total flavonoids of Wenfeihuaxian formula was (20.75 ± 2.43 )mg·ml-1. The regression equation of glucose was A = 0.0133C ± 0. 0463 ( r = 0. 999 0). The results showed that total flavonoids of Wenfeihuax-Jan formula was (154.81 ±3.00) mg·ml-1. Conclusion Ultraviolet spectrophotometry is a simple and quick approach to of Wenfeihuaxian formula with accuracy and sensitivity. The results provide a scientific basis for new drug development of Wenfeihuaxian formula.
