大黄素联合3'-叠氮-3'-脱氧胸腺核苷对人肝癌HepG2细胞增殖的协同抑制作用

摘  要:目的观察大黄素联合3'-叠氮-3'-脱氧胸腺核苷(AZT)对人肝癌细胞HepG2增殖及凋亡的影响。方法采用四唑蓝比色试验(MTT)检测大黄素、AZT及二者固定浓度比例联合在不同作用时间对HepG2细胞增殖的影响,应用Calcusyn 2.0软件计算药物半数抑制浓度(IC50)及联合用药指数(CI);流式细胞术(FCM)测定给药后细胞凋亡情况。结果大黄素与AZT联合作用比两药单用能显著抑制HepG2细胞的增殖,不同浓度的联合在不同作用时间联合指数(CI值)均小于1。联合用药组细胞凋亡率显著高于对照组和单一用药组(P<0.05)。结论大黄素/AZT联合用药具有比单用大黄素或AZT能更显著地在体外抑制HepG2细胞增殖并诱导细胞凋亡,表现出明显的协同效应。Objective To study the effects of combining emodin and 3’-azido-3’-deoxythymidine(AZT) on the proliferation and apoptosis in human hepatoma cell line HepG2 cells.Methods HepG2 cells were treated with distinct concentration of emodin,AZT and emodin combined with AZT respectively.At indicated time point,the proliferation of cells was determined by MTT assay.Morevoer,half inhibitory concentration(IC50)and combination index(CI) were calculated through Calcusyn 2.0 software.The proportion of cell apoptosis was detected by flow cytometry(FCM).Results The inhibitory rate of emodin combinated with AZT on the proliferation of HepG2 cells was significantly higher than those of emodin or AZT alone.CI values of emodin combinated with AZT not only at different concentration but also at different treated time were all less than 1.Apoptosis rate of cell in emodin combinated with AZT group was higher than that in control and emodin or AZT alone group(P〈 0.05).Conclusion Emodin combined with AZT can more potentially inhibit the proliferation and induce the apoptosis of HepG2 cells than emodin or AZT did alone,which has shown synergistic effect.
