摘 要:目的探讨绞股蓝总苷对血管性痴呆模型小鼠海马组织神经元损伤和pCREB表达的影响。方法反复夹闭两侧颈总动脉致缺血再灌注制作小鼠血管性痴呆模型。将雄性C57BL/6J小鼠80只随机分假手术(Sham)组、血管性痴呆模型(VD)组、绞股蓝总苷低、中、高剂量(100、200、400mg/kg)处理组。苏木精伊红染色法示小鼠海马组织CA1区神经元损伤的变化情况;Real-time PCR检测海马组织pCREBmRNA的表达量。结果绞股蓝总苷干预可显著减轻VD小鼠的海马神经元损伤;VD模型组海马pCREBmRNA的表达明显低于假手术组(P<0.05);绞股蓝总苷各剂量处理组海马pCREB mRNA的表达水平明显高于VD模型组(P<0.05)。结论血管性痴呆小鼠海马神经元损伤可被绞股蓝总苷减轻,上调海马神经元pCREB的表达。Objective To determined the influence of Gypenosides on the neurons injury and the expression of extracellular p-cAMP-respone element binding protein in hippocampus tissue of the mice with vascular dementia(VD).Methods The models of VD mice were established through three times ischemia—reperfusion of bilateral common carotid arteries.The male mice of C57BL/6NCrlvr were randomly separated into the sham operated group,vascular dementia model group,and gypenosides treatment group of low,middle and high dosage(100,200and 400 mg/kg) respectively.The changeon the neurons injury in hippocampal CA1 region was detected by HE staining.The expression of p-cAMP-respone element binding protein mRNA was determined by Real time PCR.Results The neurons injury in the hippocampus of vascular dementia(VD) mice could be remarkably reduced by Gypenosides treatment.The relative expression level of pCREB mRNA in the hippocampus was significantly lower in the vascular dementia model group than that in the sham operated group(P 〈 0.05).Finally,the relative expression level of pCREB mRNA in the hippocampus in low,middle and high dosage Gypenosides treatment groups were obviously higher than that in vascular dementia model group(P 〈 0.05).Conclusion Gypenosides can reduce the neurons injury in the hippocampus of VD mice,which may be relevant with the up-regulated expression of pCREB.
