摘  要:目的:探讨宫颈癌石蜡切片组织荧光原位杂交过程中不同预处理方法对实验效果的影响。方法:分别采用硫氢化钠法和亚硫酸氢钠法预处理宫颈癌石蜡组织切片,从细胞核消化程度以及荧光信号质量等方面比较不同的消化时间、变性温度以及不同年份的石蜡切片的荧光原位杂交实验效果的差异。结果:①优化后的亚硫酸氢钠法和硫氢化钠法处理石蜡切片都能得到较好的荧光原位杂交信号。②石蜡切片消化处理后以变性温度90℃处理切片,能得到高质量的荧光原位杂交信号。③两种预处理方法处理不同年份的石蜡切片得到的实验结果不同,硫氢化钠法可以处理年代更久远的石蜡切片。结论:与亚硫酸氢钠法相比,硫氢化钠法能得到质量更高的荧光原位杂交信号,在科研方面有更广阔的应用前景。Objective: To explore the impacts of different pretreatment methods of fluorescence in situ hybridization in paraffin - embedded sections of cervical cancer on experiment effect. Methods : The paraffin - embedded sections of cervical cancer were pretreated by sodium bisulfide method and sodium hydrogen sulfite method respectively; the experimental effects of fluorescence in situ hybridization under the circumstances of different digestive times, different denaturation temperatures and paraffin - embedded slices in different years were com- pared in the aspects of digestive degree of cell nuclei and fluorescence signal quality. Results: Both optimized sodium bisulfite method and sodium hydrosulfide method treated paraffin sections got better fluorescence signal. Treating paraffin slides with denaturation temperature of 90~C got high quality fluorescence signal. Both two pretreatment methods treated different years of paraffin - embedded slides got different experimental results, Sodium hydrosulfide method was proved to be more effective especially in treating more older slides. Conclusion: Compared with sodium bisulfite method, sodium hydrosulfide method can get higher quality fluorescence signals and have broad application prospects in scientific researches.
