钠葡萄糖共转运载体1在卵巢癌中的表达及作用研究

摘　　要：目的:探讨钠葡萄糖共转运载体1(SGLT1)在卵巢癌中的表达及其作用。方法:收集卵巢癌及癌旁正常组织,Western blot及PCR检测SGLT1蛋白及mRNA水平;培养类癌细胞系(carcinoid cancer)及卵巢癌细胞系(OVCAR3,SKOV3和A2780),WB及PCR检测SGLT1蛋白及mRNA水平;构建慢病毒小分子干扰SGLT1载体并筛选稳定细胞株,甲基偶氮唑蓝比色法(MTT)及BrdU掺入比色法检测细胞增殖情况,WB检测细胞周期相关蛋白的表达水平。结果:SGLT1在卵巢癌组织中表达上调,在卵巢癌细胞系OVCAR3中表达明显上调;慢病毒载体介导干扰SGLT1后,OVCAR3细胞P21及P27表达增加,p-Rb水平下调,OVCAR3细胞增殖被抑制。结论:SGLT1在卵巢癌中的表达上调,敲除SGLT1抑制OVCAR3细胞增殖。Objective： To explore the expression and effect of sodium -glucose transporter 1 （SGLT1） in ovarian cancer. Meth- ods： The ovarian cancer tissue and normal ovarian tissue adjacent to ovarian cancer were collected, Western blot and PCR were used to de- tect the levels of SGLT1 protein and mRNA; earcinoid cancer cell line and ovarian cancer cell lines （OVCAR3, SKOV3 and A2780） were cultured ; lentiviral small - molecular interfering vector was established and stable cell strains were screened, MT＇F method and BrdU incorpo- ration assay were used to detect cell proliferation, Western blot was used to detect the expression level of cell cycle - related proteins. Re- suits： SGLT1 upregulated in ovarian cancer tissue, especially in ovarian cancer OVCAR3 cells ; lentiviral vector - mediated interference of SGLT1 inhibited the growth and proliferation of OVCAR3 cells, increased the levels of P21 and P27 and reduced p - Rb. Conclusion： SGLT1 upregulates in human ovarian cancer; knockdown of SGLT1 inhibits the growth and proliferation of OVCAR3 cells.
