摘  要:目的:初步探讨CD4+T细胞表达的腺苷CD39和CD73在过敏性哮喘患者外周血中的水平变化及与叉头蛋白3(FoxP3)、GATA结合蛋白3(GATA3)和维A酸相关孤儿受体(ROR)-γt mRNA相关性。方法:选择54例对屋尘螨过敏的慢性持续期哮喘患者[间歇状态和轻度持续哮喘组(间歇-轻度组)29例,中重度组25例]和45名健康志愿者,通过荧光实时定量PCR方法检测并比较研究对象外周血CD4+T细胞中CD39、CD73、FoxP3、GATA3和ROR-γt mRNA水平。结果:CD39 mRNA水平正常对照组明显高于间歇-轻度组(P=0.000),间歇-轻度组明显高于中重度组(P=0.001)。CD73 mRNA水平正常对照组明显高于间歇-轻度组(P=0.001),哮喘亚组间差异无统计学意义(P=0.126)。哮喘组CD4+T细胞表达的CD39 mRNA与FoxP3 mRNA水平呈显著正相关性(r=0.607,P=0.000),与GATA3 mRNA水平呈显著负相关性(r=-0.505,P=0.000),与ROR-γt mRNA无显著相关性(r=-0.287,P=0.054)。哮喘组CD4+T细胞表达的CD73 mRNA与FoxP3和ROR-γt mRNA水平无显著相关性(r=0.110,P=0.450;r=-0.227,P=0.138),与GATA3mRNA水平呈显著负相关性(r=-0.286,P=0.040)。结论:CD4+T细胞表达的CD39和CD73在过敏性哮喘免疫反应过程中起到重要作用。Objective To investigate the expression of CD39 and CD73 by CD4+T cells in peripheral blood of patients with allergic asthma and its correlation with GATA binding protein (GATA)3, retinoid-ralated orphan receptor (ROR)-γt and forkhead box protein (FoxP)3 mRNA expressed by CD4+T cells. Methods Fifty-four patients with chronic persistent allergic asthma (29 with intermittent-mild form, 25 with moderate-severe form) and 45 healthy volunteers were enrolled in this study. The expression of CD39, CD73, GATA3, ROR-γt and FoxP3 mRNA in CD4+T cells was determined by fluorescent quantitative real-time PCR. Results The expression level of CD39 in CD4+T cells in healthy controls (n=45) was higher than that in intermittent-mild asthma (n=29) (P=0.000), and the level in intermittent-mild asthma was higher than that in moderate-severe asthma (n=25) (P=0.001). The expression level of CD73 in CD4+T cells in healthy controls was higher than that in intermittent-mild asthma (P=0.001), and there was no significant difference between intermittent-mild asthma and moderate-severe asthma (P=0.126). CD39 mRNA was positively correlated with FoxP3 mRNA (r=0.607, P=0.000), and negatively correlated with GATA3 mRNA (r=-0.505, P=0.000), and not correlated with ROR-γt mRNA (r=-0.287, P=0.054). CD73 mRNA was not correlated with FoxP3 and ROR-γt mRNA (r=0.110,P=0.450; r=-0.227, P=0.138), and was negatively correlated with GATA3 mRNA (r=-0.286, P=-0.040). Conclusions CD39 and CD73 expressed by CD4+T cells may play an important role in mediating the immune response in patients with allergic asthma.
