摘  要:本文探讨西洋参总皂苷(PQS)对缺血再灌注(I/R)心肌细胞凋亡的抑制机制。试验采用原代培养乳鼠心肌细胞,模拟心肌缺血-再灌注损伤模型,并给予PQS干预。应用RT-PCR、Westernblot方法和免疫荧光方法观察Bcl-2的表达,并检测细胞色素c(Cytc)的表达。结果I/R组与对照组比较Bcl-2和Cyt c的表达明显增加(P<0.05);PQS干预组较I/R组Bcl-2的表达增加,而Cyt c表达减少(P<0.05)。结论得出PQS可上调心肌细胞Bcl-2并减少Cyt c表达,从而抑制心肌细胞凋亡。To explore the possible effects of PQS on inhibiting cardiomyocyte apoptosis, the I/R cardiomyocyte model was estab- lished. The morphological alterations of apoptotic cardiomyocytes were confirmed with transmission electron microscope. The mRNA expression of bcl-2 was determined by the situ hybridization and RT-PCR, and Bcl-2 was determined by western blotting and immu- nofluorescence detection. In I/R group the evident apoptotic histopathological changes were observed and it was alleviate in PQS group. The expression levels of Bcl-2 were significantly up regulated and Cyt c decreased (P 〈0.05) in PQS group. PQS can signifi- cantly inhibit cardiomyocyte apoptosis induced by ischemia/reperfusion in rats through the increased expression of Bcl-2 and de- creased expression of Cyt c.
