乳腺癌HER2靶向分子探针的制备及体外MRI初步研究

摘　　要：目的以乳腺癌表皮生长因子受体2(HER2)为靶点,制备以顺磁性粒子钆为载体的MR分子探针,通过MR靶向成像为乳腺癌个体化治疗提供影像学依据。材料与方法利用课题组前期制备的针对HER2的荧光标记探针FITC-LTVSPWY与钆喷替酸葡甲胺(Gd-DTPA)耦联获得MR靶向探针。以表达HER2阳性的人乳腺癌细胞SKBR3、MCF-7及HER2阴性的MDA-MB-231细胞为观察对象,加入靶向探针FITC-LTVSPWY-Gd-DTPA共同孵育作为实验组,细胞加入Gd-DTPA作为对照组,不添加任何探针的细胞作为空白组,利用MRI行体外细胞成像;T1WI序列采集图像,进行视觉分析与比较各组细胞MRI信号。结果细胞体外MRI结果显示,实验组细胞T1WI呈高信号,对照组与空白组细胞均呈等信号,且存在视觉差异。结论课题组构建的以HER2为靶点的MR分子探针具有良好的磁学特性,体外可以与HER2阳性乳腺癌细胞特异性地结合,为体内成像研究奠定了基础。Purpose To prepare human epidermal growth factor receptor 2 （HER2）- targeted MR molecular probes with paramagnetic particle gadolinium as carrier, and to provide imaging data for individualized treatment of breast cancer through its targeted MR imaging. Materials and Met.hods The HER2-targeted molecular probe was made by conjugating fluorescent labeling probe FITC-LTVSPWY and Gd-DTPA prepared by our research group earlier. Positive expressions like human breast cancer cell SKBR3 and MCF-7 and negative expression like MDA-MB-231 were selected as observe objects. Those cells incubated with FITC-LTVSPWY-Gd-DTPA were taken as experimental group, those with Gd-DTPA as control group, and those without any probe as blank control group. Then MRI was performed in vitro and TlWI signal of cells was collected and assessed visually. Results The strong signals on TIWI were observed in the experimental group, the control group and the blank control group both showed iso-intense signals, and visual difference could be detected. Conclusion The HER2-targeted MR probe prepared by our research group shows good magnetic features. It can target HER2 in the detection of breast cancer cells in vitro, which can be used in further research on imaging in vivo.
