摘 要:目的观察2型糖尿病(T2DM)大鼠胰腺中尾加压素Ⅱ(UⅡ)及GPR14的表达,探讨UⅡ及GPR14与T2DM大鼠糖脂及胰岛素代谢水平的相关性。方法将28只SD大鼠随机分为NC组和DM组,每组14只,分别予普通和高脂饲料喂养。8周末,DM组按25 mg/kg腹腔注射链脲佐菌素溶液造模,NC组注射同体积无菌柠檬酸钠溶液作对照,10周末行OGTT筛选成模大鼠。第4、8、14周末分别取血检测UⅡ、糖脂及胰岛素相关指标。14周末处死所有大鼠,胰腺组织用HE染色并在光镜下观察病理改变,用免疫组化SP及实时荧光定量RT-PCR法检测UⅡ及GPR14在胰腺的表达,ELISA法检测血清UⅡ水平。结果 8周末DM组UⅡ、空腹血糖(FPG)、空腹胰岛素(FINS)、TG、TC均高于同周NC组(均P<0.001),HDL-C、胰岛素敏感指数均低于同周NC组(均P<0.05),但14周末FINS与8周末相比已下降。14周末,2组UⅡ及GPR14在胰腺表达比较,DM组明显增高(P<0.001)。Pearson相关分析示,UⅡ及GPR14含量与FPG、TG和TC呈正相关(均P<0.05),与ISI和HDL-C呈负相关(均P<0.05)。结论 T2DM大鼠胰腺组织高表达UⅡ及GPR14,提示其可能与T2DM发病相关。Objective To observe the expression of urotensin Ⅱ( UⅡ) and its G protein-coupled receptor-14 (GPR14) in the pancreas of rats with type 2 diabetes (T2DM), and further to study the correlation of UⅡ-GPR14 system and the metabolism of glucose , adipose and insulin in T2DM rats.Methods A total of 28 male Sprague -Dawley rats were randomly divided into two groups according to random number table :normal control group ( Group NC) and diabetes mellitus group (Group DM);in which normal and high -fat diets were fed to the subjects, respectively.At the end of 8^th week, intraperitoneal injection of STZ by 25 mg/kg was given to Group DM, while sterile sodium citrate was given to Group NC as control.At the end of 10th week, OGTT was used for selection of model rats.At the end of the 4th, 8^th and 14^th week, UⅡand the relevant metabolic indicators of glucose , adipose and insulin were measured .At the end of the 14^th week, all the rats were sacrificed for collection of pancreatic tissue and pathological observation .The expression of UⅡand GPR14 in pancreatic tissues were assessed by immunohistochemical stretavidin proxidase ( SP) method and realtime fluorescent quantitative reverse transcription -polymerase chain reaction ( RT-PCR).Results At the end of the 8th week, the UⅡ, FPG, FINS, TG and TC in Group DM were significantly higher than those in Group NC (P〈0.001), while HDL-C and ISI were significantly lower in Group DM (P〈0.05);however, FINS at the end of 14th week was lower than that at the end of the 8th week.At the end of the 14th week, the expressions of UⅡand GPR14 in Group DM were significantly up-regulated ( P〈0.001) .Pearson linear correlation analysis shown that pancreatic UⅡ and GPR14 were significantly positively correlated with FPG , TG and TC ( P〈0.05 ) , and negatively correlated with ISI and HDL -C (P〈0.05).Conclusion The high expression of UⅡand GPR14 in the pancreas of rats with T2DM is correlated to the pathogenesis of T2DM.
