摘 要:连翘是最广泛使用的传统中药之一,具有抗菌、抗病毒、抗炎和抗氧化活性等重要的药理作用,而连翘苷是连翘的主要生物活性成分。为了从连翘叶中快速简便地提取、分离和纯化得到纯度较高的连翘苷,试验采用无水乙醇和体积浓度85%乙醇回流提取,大孔树脂柱层析和石油醚萃取的方法分离,重结晶法进行纯化,经不同组合试验以确定最佳制备工艺。通过HPLC测定比较所得连翘苷的含量、纯度,最终确定了以无水乙醇回流提取,石油醚萃取分离,重结晶纯化的工艺流程,通过该方法可以得到纯度达90%的连翘苷。Forsythia suspensa is one of most widely used traditional Chinese medicines, and possesses important biolog-ical activities, including antibacterial, antiviral, anti-inflammatory and antioxidant activities. Forsythin is the main bioac-tive component of forsythia suspensa. This paper aimed to obtain highly purified forsythin from forsythia suspensa leaves quickly and easily, and to study its pharmacological effects in future. The study designed a series of methods, extraction methods containing anhydrous ethanol and 85% ethanol extract, separation methods adopting macroporous resin column chromatography and petroleum ether extraction, purification method employing recrystallization. Through HPLC deter-mined the content and purity of forsythin, a better method was founded which included anhydrous ethanol extraction, sepa-ration of petroleum ether and recrystallization. At last, a certain amount of 90% purity forsythin had been extracted by the method.
