摘  要:目的探讨WT1基因在慢性粒细胞白血病中的表达与临床意义。方法采用Taqman荧光探针逆转录-聚合酶链反应(RT-PCR)检测慢性粒细胞白血病(chronic myelocytic leukemia,CML)患者38例(其中慢性期25例,进展期13例)和10例正常健康人外周血细胞WT1基因的表达量,并进行分析。结果 10例正常人均未检测到WT1基因的表达,38例CML患者22例表达阳性(57.8%),慢性期12例表达阳性(48.0%),进展期10例表达阳性(76.9%)。WT1基因在CML中高表达,WT1基因的表达与疾病的进展呈正相关。WT1基因的表达量与外周血原始细胞的百分比成正相关。结论检测WT1基因的表达可作为评估CML风险的重要指标之一。Objective To explore the WT1 gene expression and its clinical significance in chronic myelocytic leukemia (CML). Methods WT1 gene expression in peripheral blood of 38 CML patients(25 in chronic phase and 13 in progressive stage) and 10 normal patients were detected and analyzed by reverse transcription-polymerase chain reaction (RT-PCR). Results There were 22 patients in CML(57. 8% ) that with WT1 gene positive expression, which was not found in 10 normal patients. The 22 positive patients included 12 in chronic phase (48.0%) and 10 in progressive stage(76. 9%). WT1 gene got higher expression in CML compared with the normal patients. WT1 gene expression positively correlated with the progress of CML and the expression level positively correlated with the percentage of peripheral blood progenitor cells. Conclusion WT1 gene expression might be used as one of the important indexes to assess the risk of CML.
