摘  要:目的研究地塞米松诱导小鼠急性药物性脂肪肝模型是否具有自愈性,了解其自愈性及其特征,探讨该疾病动物模型在脂肪肝研究领域的适用性。方法腹腔注射地塞米松(100mg/kg×4d)辅以高脂饮食建立小鼠急性药物性脂肪肝模型,正常对照组0.9%氯化钠溶液腹腔注射;分别于末次注射后1、2、3、4d分批次取材测定肝指数、肝组织粗脂肪含量,并进行病理组织学分析;生化检测血清总胆固醇(TC)、三酰甘油(TG)含量。结果地塞米松辅以高脂饮食可成功建立小鼠急性药物性脂肪肝模型,在造模干预因素撤除后,升高的肝指数、肝组织粗脂肪含量及血清TC、TG等指标呈进行性下降并趋于正常。结论地塞米松诱导小鼠急性药物性脂肪肝模型可在数日内自愈,因此该模型不可用于脂肪肝防治药物的筛选。Objective To investigate whether the dexamethasone induced mice acute fatty liver model has self-healing,to realize its self-healing and feature,and to explore the applicability of fatty liver animal model in the field of fatty liver.Methods Acute drug-induced fatty liver model was established by peritoneal injection of dexamethasone(100mg/kg × 4d)combined with high-fat diet,and a normal control group was set up by peritoneal injection of 0.9%sodium chloride at the same time;then the materials in batch 1,2,3,4dwere drown after the last injection,respectively;liver index and liver crude fat content were determined;histopathological analysis was performed;and total cholesterol(TC)and triglyceride(TG)level were detected.Results The dexamethasone combined with high-fat diet can successfully establish a mouse model of acute drug-induced fatty liver,but after the modeling intervention factor was removed,the elevated liver index,liver crude fat content and serum TC,TG and other indicators decreased progressively and even became normal.Conclusion Dexamethasone induced mice acute fatty liver model can heal within a few days,therefore,it can not be used to sieve the drug for prevention and treatment of fatty liver.
