摘  要:目的探讨胱硫醚γ-裂解酶在缺氧缺血性脑病(HIE)新生大鼠肾脏肾细胞凋亡中的作用机制。方法新生7 d Wistar大鼠制备HIE模型。分为假手术对照组和HIE组,于HIE后6 h、12 h、24 h和48 h处死大鼠,取肾脏组织,应用流式细胞术检测肾细胞凋亡情况;MPO检测肾脏组织炎性损伤的程度;应用免疫组织化学法检测肾细胞Bax蛋白的表达情况;应用Western blot技术检测细胞凋亡与p-JNK信号通路的关系。结果 HIE组6 h肾脏阳性凋亡细胞开始增多,24 h达到高峰,各时间点阳性凋亡细胞均高于对照组(P<0.05);HIE组6 h Bax阳性细胞表达开始增多,24 h达到高峰,各时间点阳性细胞均高于对照组(P<0.05);HIE组6 h MPO的OD值增高,24 h达到高峰,各时间点OD值均高于对照组(P<0.05);HIE组6 hp-JNK蛋白表达开始增多并24 h达到高峰;对照组CSE蛋白均高于HIE组(P<0.05),CSE随时间逐渐降低。结论 HIE新生大鼠肾脏细胞CSE蛋白表达降低,对肾脏细胞凋亡有抑制作用,其可能通过p-JNK信号通路实现的。Objective To investigate the effect of cystathionine ν lyase on apoptosis of renal cells of neonatal rats with hypoxic-ischemic encephalopathy ( HIE), and to explore its action mechanism. Methods The Wistar rats (7 - day age) were prepared as HIE model, which were randomly divided into sham - operation control group and HIE group. The rates were executed at 6h, 12h,24h,48h after HIE to obtain kidney tissues. The apoptosis condition of renal cells was detected by flow cytometry,the degree of inflammatory injury of renal tissue was observed by MPO, Bax protein expression was determined by immunohistochemistry, and the correlation between renal cell apoptosis and p-JNK signaling pathway was analyzed by Western Blot. Results In HIE group,positive apoptosis cells were increased at 6h and peaked at 24h, and the positive apoptosis cells in different time points were significantly higher than those in control group ( P 〉0.05 ). The OD value of MPO in HIE group was increased at 6h,which reached peak at 24h,and the OD value in different time points was significantly higher than that in control group ( P 〈0.05 ). The expression levels of p-JNK protein were increased at 6h and peaked at 24h. The expression levels of CSE protein in control group were significantly higher than those in HIE group ( P 〈0.05), furthermore,which were gradually decreased with the time going on. Conclusion The expression levels of CSE in renal cells of neonatal rats with HIE are decreased, which has inhibitory effect on renal cell apoptosis, which may be realized through p-JNK signaling pathway.
