摘  要:目的探讨人参皂苷Rb1对癫痫大鼠学习记忆及海马kalirin-7和突触后致密物95(PSD-95)表达的影响。方法 40只SD大鼠随机分为4组:对照组、模型组、Rb1低、高剂量组。模型制备成功后,采用Morris水迷宫检测其行为学,电镜法观察海马CA1区突触超微结构,Western blot法检测海马kalirin-7和PSD-95的表达。结果模型组大鼠的末次逃避潜伏期、穿台次数、海马CA1区PSD厚度、kalirin-7和PSD-95表达[分别为(11.2±1.6)s、(4.9±1.3)次、(25.9±2.4)nm、(0.32±0.03)、(0.95±0.08)];与对照组[分别为(4.5±0.8)s、(10.9±2.1)次、(34.1±3.7)nm、(0.86±0.07)、(1.94±0.13)],比较均具有显著性差异(P<0.05)。而人参皂苷Rb1干预组剂量依赖性地逆转了上述的异常变化(P<0.05)。结论人参皂苷Rb1可能通过上调海马kalirin-7和PSD-95蛋白的表达来改善癫痫大鼠的认知功能障碍。Objective To explore the effects of ginsenoside on behaviors and expression of synaptic associated proteins in the hippoeampus of epileptic rats. Methods Forty rats were randomly divided into 4 groups : control, epileptic,low dose of ginsenoside Rbl and high dose of ginsenoside Rbl groups. Rats were given PTZ till being kindled successfully. Their behaviors were afterwards determined by Morris water maze, morphology was observed by electron microscope, and expressions of kalirin-7 and PSD-95 in the hippocampus were detected by Western blot. Results Compared to the control group, the ability of learning and memory was impaired in the escape latency ( 11.2 +- 1.6 compared to 4.5 +- 0.8 ) and in the number of crossing (4.9 -+ 1.3 compared to 10.9 -+ 2.1 ) ( P 〈 0.05 ), the synaptie uhrastructure of hippocampus CA1 showed significantly changes in the PSD-95 thickness (25.9 ± 2.4 compared to 34.1 ± 3.7 ) ( P 〈 0.05 ), and expressions of kalirin-7 (0.32 ± 0.03 compared to 0.86 ±0.07) and PSD-95 (0.95± 0.08 compared to 1.94 ± 0.13 ) obviously decreased in rats with PTZ-kindled epilepsy ( P 〈 0.05 ) , while ginsenoside Rbl could dose-dependently reverse such abnor- mal changes (P 〈 0.05 ). Conclusion Ginsenoside Rbl could effectively improve the cognitive impairment of rats with PTZ-kindled epilepsy by up-regulating the expressions of kalirin-7 and PSD-95 in the hippocampus.
