摘 要:目的研究大承气汤在促进胃肠动力中的作用及其机制。方法采用电生理学、免疫荧光化学测定正常对照组、生理盐水组、大承气汤药物干预组及大承气汤+ZD7288阻断剂组大鼠胃肠电活动情况及胃肠组织中HCN1(hyperpolarizationactivated cyclic nucleotide-gated cation nonselective channel 1)的表达;给予HCN1特异性阻断剂ZD7288,检测上述指标的变化并进行分析比较。结果正常组大鼠和生理盐水组大鼠的胃肠MMC及HCN1的表达均无统计学差异;而使用ZD7288干预组后,上述指标均有显著性变化(P<0.01)。结论大承气汤能显著地促进大鼠的胃肠运动,离子通道HCN1可能是其重要的作用靶点。Objective To explore the promotive effects of Dachengqi decoction on gastrointestinal motility and its mechanism. Methods The rats were randomized into four groups:normal group, normal saline group, Dachengqi decoction group and Dachengqi de- coction + ZD7288 group. The electrophysiology and immunofluorescence stain were used to detect the gastrointestinal electrical activity and the hyperpolarization-activated cyclic nucleotide-gated cation non-selective channel 1 ( HCN1 ) expression in gastrointestinal tis- sues. Results Compared with normal group and normal saline group,the expression of gastriointestinal MMC and HCNI was significantly different in Dachengqi decoction group. Compared with Dachengqi decoction group, the expression of gastriointestinal MMC and HCN1 were significantly decreased in ZD7288 group ( P 〈 0. 01 ). Conclusion Dachengqi decoction may promote gastrointestinal motility obviously through regulating HCN1 which may be the target ion channel of Dachengqi decoction.
