摘 要:为寻找评价鸭科属间或品种间种质特性以及有效区分不同品种的工具,选取北京鸭、金定鸭、绿头野鸭、斑嘴鸭和番鸭共计40个个体,扩增其mtDNA D-loop区序列,最终获得家鸭616 bp有效序列,检测到6处插入/缺失,114次颠换,94次转换;获得番鸭619 bp有效序列,发现1处插入/缺失,4次转换,未发现颠换。利用MEGA5.0和DnaSP V.5.0等软件进行碱基组成、位点突变和单倍型分析。结果表明:鸭mtDNA D-loop区的碱基C的含量最高,并表现出了碱基组成的偏倚性;共检测到21种单倍型,Tajima’s D值为-0.11344,符合中性突变,绿头野鸭和北京鸭的核苷酸多样度较高,遗传多样性丰富;番鸭的单倍型多样度(Hd=0.429)和核苷酸多样度(Pi=0.00139)均最低,其遗传多样性相对较低。In order to seek tools for evaluating the breed characteristic among intergeneric and distinguishing different breeds, Beijing, Jinding, Lvtou, Banzui, and Muscovy duck (in total 40)were selected to amplifier the sequences of mtDNA D-loop. 616 bp sequence of domestic duck, 619 bp sequence of muscovy duck was obtained, respectively. In domestic ducks, 6 insertion/deletion, 114 times transversion and 94 times transition were detected, and in Muscovy duck, only 1 insertion/deletion and 4 times transition were found, but no transversion was detected. MEGA5. 0 and DnaSP V. 5.0 were used to compute the base compositions, mutations and haplotypes. The results showed that the mtDNA D-loop had the highest amount of base C, and showed the base composition of bias. 21 haplotypes were detected in all, and the Tajima's D value was -0.11344 which suggested that all mutations were in line with the neutral mutation. The nucleotide diversity (Pi) of Lvtou and Beijing duck were higher suggested abundant genetic diversity, the haplotype diversity (Hd=0.429)and nucleotide diversity (Pi=0.00139)in Muscovy duck were the lowest which declared low genetic diversity.
