摘 要:研究L-肉碱对氧化应激veto细胞增殖及抗氧化功能的保护作用。以vero细胞为研究对象.采用四甲基偶氮唑盐(MTT)法测定不同浓度H2O2(0.1、1、10、100、1000μmol/L)和L-肉碱(0.001、0.01、0.1、1、10、100mmol/L)预处理对vero细胞活力的作用。采用生物化学方法检测L-肉碱预处理对氧化应激vero细胞及上清液中抗氧化酶的影响。结果表明:100μmol/L和1000μmol/LH2O2显著降低vero细胞活力(P〈0.05);0.1mmol/L和1mmol/L L-肉碱预处理6h组细胞活力与对照组无显著性差异(P〉0.05)。与氧化应激组相比。0.1mmol/L和1mmol/L L-肉碱组细胞上清液中丙二醛(MDA)含量明显降低(P〈0.05),谷胱甘肽过氧化物酶(GSH—Px)、超氧化物歧化酶(SOD)活性及总谷胱甘肽(T—GSH)、还原性谷胱甘肽(GSH)和总抗氧化能力(T—AOC)水平均显著升高(P〈0.05)。0.1mmol/L和1mmol/L L-肉碱能显著提高氧化应激细胞中T—AOC和T—GSH水平(P〈0.05).显著提高谷胱甘肽过氧化氢酶(GR)、过氧化氢酶(CAT)、GSH—Px和SOD酶活性(P〈0.05)。本试验表明,0.1—1mmol/L L-肉碱可提高vero细胞的抗氧化能力,对veto细胞抗H2O2氧化应激有较好的保护作用。Protective effects of L-carnitine on cell viability and antioxidant function of vero cells were investigated in this study. Using vero cells as study object in vitro culture, the effects of defferent concentrations of H2O2 (0.1, 1, 10, 100, 1 000 μmol/L)and L- carnitine (0.001, 0.01, 0.1, 1, 10, 100 mmol/L)on cells viabilies of vero cells were measured by methyl thiazolyl tetrazolium(MTT)method; the content of antioxidants and activities of antioxidant enzymes in supernatant and oxidative stress cells were determined by biochemical methods. The results showed that concentration of H2O2 with 100 and 1000 μmol/L could significantly decreas viabilities of vero cells (P〈0.05). There were no significant differences in cell viabilities between the pretreatment groups with 0.1 and 1 mmol/L L-carnitine for 6 h and the control group (P〉0.05). Compared to the oxidative stress group, 0.1andl mmol/L L-carnitine could significantly reduced the malondialdehyde (MDA) content (P〈0.05) and significantly improve enzyme activities of superoxide dismutase (SOD), glutathione peroxidase (GSH-Px) , and the content of total glutathione (T-GSH), reductive glutathione (GSH), total antioxidant capacity (T-AOC) in supernatant of oxidative stress cells (P〈0.05). The content of T-GSH, GSH and enzyme activities of glutathione peptide catalase (GR), catalase (CAT), GSH-Px and SOD were significantly enhanced in 0.1 and 1 mmol/L L-carnitine groups (P〈0.05). It is concluded that 0.1-1 mmol/L L-carnitine can enhance antioxidant capability of the damaged vero cells, L-carnitine has better protective effect on vero cells against H2O2 oxidative stress.
