摘 要:目的:建立头孢他啶的阻抑荧光动力学测定方法。方法:利用头孢他啶对罗丹明 B-KBrO3体系的荧光反猝灭作用,以559.0 nm 为激发波长,581.0 nm 为发射波长,以对应试剂空白的荧光强度为F0,计算△F=F-F0。根据△F值与溶液中头孢他啶的浓度呈正比关系,建立阻抑荧光动力学法测定头孢他啶的新方法。结果:头孢他啶的质量浓度在0.636-8.268 ng/mL 范围内与其荧光强度呈良好线性关系(R=0.9998),检出限可达0.283 ng/mL。结论:该法简便、快速、可靠,可用于药物制剂中头孢他啶含量的测定。Objective:To develop a kinetic-fluorimetric method for the determination of ceftazidime.Methods:The method is based on the anti-fluorescence quenching effect on rhodamine B-KBrO3 .559.0 nm and 581.0 nm were chosen as the excitation and emission wavelength for measurement of the fluorescence intensity difference △F=F- F0 of blank solution and test solution,and according to positive correlation between the value of △F and the concentration of ceftazidime.A new method of anti-fluorescence quenching for the determination of ceftazidime was established.Results:The linear range of the determination is 0.636-8.268 ng/mL(R=0.9998)with the detection limit of 0.283 ng/mL.Conclusion:The method is simple,rapid and credible,which can be used for the determination of ceftazidime in pharmaceutical formulation.
