摘 要:目的:检测丰富环境对孕期应激大鼠子代海马糖皮质激素受体(GlucocorticoidReceptor,GR)的影响。方法:SD孕鼠随机分为2组,正常组和实验组。正常组动物不做任何处理,其子代断乳后在标准环境下喂养;实验组动物在孕第13~19天接受限制性应激,其子代断乳后一半在标准环境下喂养(构成应激组),另一半子代在丰富的环境下喂养(构成应激+丰富环境组)。所有子代出生后第35天被处死。用ABC免疫组织化学和Westernblot方法分别检测海马GR的表达。结果:GR阳性产物广泛分布于海马各区神经元,应激组GR表达明显少于正常组;应激+丰富环境组海马GR的表达明显多于应激组,与正常组相比差别无统计学意义。结论:丰富环境能有效逆转孕期应激造成的子代海马GR的表达降低。Objective: To observe the effects of enriched enviroment on the hippocampal glucocorticoid receptor of antenatal stressed offsprings. Methods: Pregnant SD rats were randomly separated into normal and experimental groups. The normal group accepted no stress. After weaning at postnatal day 21, their offsprings were given at the standard enviroment. The experimental group accepted the restraint stress at pregnancy day 13- 19. The half of offsprings in experimental group were given the standard enviroment (stressed group), the other part was given the enriched enviroment treatment at postnatal day 22 - 35 (stress +enriched enviroment group). The GR expression in the hippocampus was observed by immunohistochemistry staining and western blot. Results:The expression of GR were found in the hippoeampus of SD rats. Compared to the normal group,the GR expression of hippocampus in stressed group was meaningfully decreased. The GR expression of hippocampus in stress + enriched enviroment group was meaningfully higher than that of stressed group. Conclusion: Enriched enviroment can efficiently reverse the reduction of hippocampal GR expression in offsprings caused by antenatal stress.
