摘  要:目的探究青蒿琥酯对实验性自身免疫性重症肌无力(experimental autoimmune myasthenia gravis,EAMG)大鼠TNF-α、IgG2a、IgG2b的影响。方法用人工合成的大鼠来源的乙酰胆碱受体(acetylcholine receptor,AChR)α亚基97-116肽段免疫Lewis大鼠制造重症肌无力(myasthenia gravis,MG)模型,随机分为青蒿琥酯大剂量组(100mg/kg)、小剂量组(10mg/kg)和模型对照组,流式细胞仪检测不同组大鼠淋巴结单个核细胞(mononuclear cell,MNC)中TNF-α+细胞的百分比;酶联免疫吸附法(ELISA)检测EAMG大鼠血清中抗R97-116IgG2a和IgG2b抗体的水平。结果与对照组相比,青蒿琥酯小剂量组明显下调了TNF-α+的百分比(P=0.046),而大剂量组中TNF-α下调不显著,两治疗组血清中抗R97-116IgG2a和IgG2b抗体的水平下降,且小剂量组血清中抗R97-116IgG2a和IgG2b抗体的水平下降更显著。结论青蒿琥酯主要是通过下调TNF-α+细胞的数量和降低血清中抗R97-116IgG2a和IgG2b抗体水平来改善EAMG大鼠的病情。Objective To investigate the effect of artesunate on TNF-α,IgG2a,IgG2b of experimental autoim- mune myasthenia gravis (EAMG) in rats. Methods EAMG can be induced by subcutaneous injection into both hind footpads with a synthetic peptide corresponding to region 97-116 of the rat AChRα subunit (R97-116 peptide). Artesunate was administered by gavage to EAMG rats. The percentage of TNF-α+ cells among lymph nodes (mononuclear cell,MNC) was analyzed by flow cytometry. Anti-R97-116 peptide IgG2a and IgG2b antibodies in serum were detected by ELISA. Results On the clay of the experiment termination,the rats in 10 mg/kg artesunate-treated group exhibited less percentage of TNF-α+ cells and lower level of anti-R97-116 IgG antibodies (P〈0.05) com- pared with rats in control group. The therapeutic effect of artesunate on EAMG rats was associated with down-reg- ulated TNF-α+ cells (P〈0.05) and decreased anti-R97-116 IgG antibody (P〈0.01). Conelusion Artesunate could ameliorate EAMG by down-regulating the number of TNF-α+ cells and decreasing anti-R97-116 IgG antibody.
