摘　　要：[目的]探讨人骨髓核分化抗原(MNDA)对人骨肉瘤Saos-2细胞增殖及侵袭的影响。[方法]逆转录聚合酶链反应(RT-PCR)法定量测定Saos-2细胞中MNDA的mRNA表达差异。将MNDA基因转染Saos-2细胞,MTT法测定检测细胞增殖程度的改变,Transwell小室法测定肿瘤细胞侵袭能力的改变。没有转染质粒的和转染空质粒的Saos-2做为对照组。[结果]MNDA的mRNA在骨肉瘤Saos-2细胞中表达明显降低。转染MNDA的人骨肉瘤Saos-2细胞的增殖率低于对照组,并且抑制能力随时间延长抑制而增加,侵袭穿透能力减弱。[结论]人骨肉瘤转染MNDA可抑制肿瘤细胞的增殖,降低肿瘤细胞的侵袭穿透能力。[ Objective] To detect the influence of MNDA gene in the proliferation and invasion of osteosarcoma cell line Saos -2. [ Methods] Osteosarcoma cell line Saos -2 was cultured in DMEM medium. RT -PCR was performed to compare the differences in the expression of MNDA mRNA in the cell lines. Then, MNDA was transfected into the cell lines. MTT assay was used to detect the changes in the cell proliferation rate after enhancing MNDA expression by transfeetion, and invasion of tumor ceils was measured by using transwell chamber assay. Osteosarcoma cell line Saos -2 without transfeetion was used as the con- trol. [Results] Compared to the control group, the expression of MNDA mRNA was significantly lower in the Saos-2 cell line. The proliferation rate was significantly lower in the transfected Saos - 2 than in the control group, and the invasion of Saos -2 ceils was significantly decreased in the transfected MNDA group. [ Conclusion] The results suggest that MNDA could pro- mote cellular apoptosis, decrease cell proliferation, and inhibit the invasion of osteosareoma cells.
