单核细胞^(18)F-FDG标记与蛛网膜下腔示踪研究

摘  要:目的:^(18)F-FDG体外标记外周血单核细胞,异体植入正常新西兰兔蛛网膜下腔,观察其演变情况以及对动物的整体影响。方法:经兔耳动脉抽取外周血,分离单核细胞,用放射性核素^(18)F-FDG标记,经枕大池植入蛛网膜下腔,分别于植入细胞后15min、1.5h以及3h进行正电子发射型计算机断层显像(PET)/CT显像。显像后观察兔子体温、饮食和活动情况。结果:单核细胞^(18)F-FDG的标记效率(经四次清洗后)达到92.42%。PET/CT显像发现,植入细胞会快速地沿着蛛网膜下腔向头端(达嗅脑水平)和尾端扩散(最下端在上腰椎水平,放射性核素的浓集程度逐渐减弱,实际扩散的距离达19cm),且蛛网膜下腔中的放射性分布清楚持久。兔子体温在正常范围内波动,饮食、活动量没有显著性改变。结论:利用^(18)F-FDG标记单核细胞蛛网膜下腔示踪,是在活体上有效的整体监测植入细胞的办法。植入细胞迅速沿蛛网膜下腔广泛分布的特点,应该在制定细胞移植策略和疗效预测时给予充分考虑。异体外周血单核细胞注入蛛网膜下腔后对兔子整体没有明显的影响。Objective: To implant allograft in monocytes labelled with 18F-FDG in vitro subarachnoid space of normal New Zealand rabbit, the evolvement of the implanted cells and the host's state were monitored. Methods: The monocytes were isolated from peripheral blood drawn from the rabbit ear artery, then the mono- cytes were labelled with 18F-FDG in vitro and then injected into subarachnoid space ria cistema magna. PET/CT scan was performed in 15 min, 1.5 h and 3 h after implant. The temperature, diet and activity of the rabbit were observed after the cells injected. Results: After being washed four times, the cell labelling efficiency of lSF-FDG reached 92.42%. The PET/CT imaging showed that implanted cells rapid diffused along the subarachnoid space to the head direction (reached at the olfactory brain levels) and the tail direction (reached the lower end of the lumbar level). The concentration of radionuclides decreased gradually and the diffusion distance was up to 19 cm. Moreover, the distribution of radioactivity in the subarachnoid space was clear and lasting a long time. The temperature of rabbit was fluctuated within the normal range, and the diet, activity levels did not change significantly. Conclusion: Tracing monocytes labelled with 18F-FDG with PET/CT scan is an effective method to monitor implanted cells in vivo in the subarachnoid space. The characteristics of implanted cells rapidly distributed along the subarachnoid space contribute to the strategies and prediction of the cell transplantation treatment. The rabbit doesn't affect after intrathecal injection of allogous monocvtes into subarachnoid space.
