乙酰葛根素对Aβ_(25-35)诱导BV-2小胶质细胞形态及TNF-α的影响

摘　　要：目的研究乙酰葛根素对Aβ_(25-35)诱导的BV-2小胶质细胞的形态、TNF-α蛋白及基因表达的影响。方法将BV-2小胶质细胞进行培养,用Aβ_(25-35)诱导小胶质细胞建立细胞模型,加入不同浓度的乙酰葛根素,使用倒置相差显微镜观察BV-2小胶质细胞的形态、Western blot检测TNF-α蛋白表达水平、荧光定量PCR检测TNF-αmRNA表达。结果乙酰葛根素可以抑制Aβ25-35诱导的BV-2小胶质细胞的激活,降低TNF-α蛋白及mRNA的表达(P<0.05)。结论乙酰葛根素对Aβ_(25-35)诱导的BV-2小胶质细胞可以发挥一定的保护作用,本研究为阿尔茨海默病(AD)的药物治疗提供了实验依据。Objective To determine the effect of Acetylpuerarin on the expression of TNF-αprotein and mRNA in BV-2 microglia cell induced by Aβ25-35 ,and observe the cell morphology .Methods BV-2 microglia cells were cultured under routine condition ,treated with Aβ25-35 and Acetylpuerarin when it reached 70%~80%.Then we used inverted phase contrast microscope for cell morphology ,Western blot assay for the protein expression of TNF-αand real-time PCR assay for the mRNA expression of TNF-α.Results Acetylpuerarin could inhibit the activation of BV-2 microglia cells induced by Aβ25-35 and attenuate the mRNA and protein expression of TNF-α（P〈0.05）.Conclusion Our founding suggest that Acetylpuerarin may play a protective role in BV -2 microglia cells induced by Aβ25-35 ,and this will provide experimental basis for the medication of Alzheimer＇ s disease.
