摘  要:目的利用荧光PCR检测技术和方阵原理相结合,建立方阵式荧光PCR检测从业人员肠道致病菌的方法。方法采集从业人员肛拭子样品经增菌培养后,把待检样品排成方阵,对方阵中的每排、每列样品分别混合为一个新的样品,对混合样品进行沙门菌、志贺菌双色荧光PCR检测,检出的疑似阳性样品用传统的分离培养法进行培养鉴定。结果荧光PCR与方阵相结合,比传统培养法灵敏度高、特异性强,而且能快速准确定位疑似阳性样品,操作简便快捷。结论方阵式荧光PCR快速、灵敏度高、特异性强,可提高从业人员带菌检出率并大大降低工作强度,此方法可用于从业人员体检的快速诊断和肠道传染病的初筛,同时值得在大样本量的检测工作中推广。Objective To establish a method for detection of pathogenic enterobacteria in practitioners by combination of fluorescence PCR detection technology and square matrix principle.Methods Anal swab samples were collected from employees for enrichment culture,then the samples under test were arranged in square array.Samples of each row and each column were mixed into a new sample separately to detect the Salmonella and Shigella by using dual-color fluorescence PCR.The suspected positive samples were cultured and identified with the traditional separation culture method.Results Combination of fluorescence PCR and square matrix showed higher sensitivity and strong specificity compared with traditional culture method,and it can quickly and accurately detect the suspected positive samples,convenient and simple.Conclusion Combination of fluorescence PCR and square matrix can improve the bacteria detection rate and reduce work load at the same time.With rapidity,high sensitivity and strong specificity,it could be used for rapid diagnosis of employees in physical examination and early screening of intestinal infectious diseases,worth popularizing in detection of large samples.
