中国汉族人及云南省彝族、傣族人GPIbα基因VNTR多态性研究

摘　　要：目的　研究中国汉族人及云南省彝族、傣族人血小板膜糖蛋白 (GP)Ibα基因可变数目串联重复序列 (VNTR)的多态性特点。方法　应用聚合酶链反应 (PCR)技术 ,聚丙烯酰胺凝胶电泳 ,溴乙锭染色 ,PCR产物测序分析 ,研究 110名汉族人、47名彝族人和 41名傣族人等位基因的杂合情况。结果　在中国汉族人、云南省彝族及傣族人群中 ,39bp的VNTR只出现一次重复 (D)和二次重复 (C)二种等位基因 ,而未发现三次重复 (B)和四次重复 (A)等位基因 ,其C等位基因和D等位基因的频率分别为 :汉族人 0 5 3∶0 47、彝族人 0 6 6∶0 34、傣族人 0 5 4∶0 46 ,总的理论杂合率分别为 :汉族人 49 8%、彝族人 44 9%、傣族人 49 8%。结论　中国汉、彝、傣三个民族人群中GPIbα基因VN TR多态性的表型或其等位基因频率无明显差异 。Objective To study the variable number of tandem repeat(VNTR) polymorphism of platelet glycoprotein(GP) Ibα gene in Han,Yi and Dai populations.Methods About 437 bp amplified fragment of the human GPIbα gene from 110 unrelated individuals of Han group and 47 of Yi group and 41 of Dai group were analyzed by 8% polyacrylamide gel with ethidium bromide staining.PCR product was sequenced.Results The D variant (with 1 repeat) and the C variant(with 2 repeats) of the VNTR of the 39 bp sequence were observed in the three groups.The larger variants containing three or four tandem repeats weren′t found in those populations.The C/D allele frequencies were 0 53∶0 47 in Han group,0 66∶0 34 in Yi group and 0 54∶0 46 in Dai group,respectively.The theoretical heterozygous rates were 49 8% in Han,44 9% in Yi and 49 8% in Dai population,respectively.Conclusion The VNTR polymorphism of GPIbα gene in Han,Yi and Dai populations was very different from that observed in the European caucasians.
