摘 要:目的建立一种肉中志贺氏菌的快速检测方法。方法利用FTA(Flinders Technology Associates)滤膜从肉中提取志贺氏菌基因组DNA,以志贺氏菌属的ipaH基因为模板设计引物进行PCR反应得到369 bp片段,经测序证实该片段为目的扩增产物。结果采用建立的FTA滤膜结合PCR检测肉中志贺氏菌的方法,人工污染牛肉、羊肉、猪肉、鸡肉匀浆液的检出限均为8×101CFU/ml匀浆,6 h即可完成整个检测过程。结论该方法灵敏度高、耗时短,为预防志贺氏菌食物中毒、控制细菌性痢疾的暴发流行提供了新的检测方法。Objective To establish a rapid method for detection of Shigella in meat. Methods By using the FTA( Flinders Technology Associates) to extract the Shigella genome DNA within the meat,the plasmid( ipaH)gene was used as template to design primer PCR reaction to get A DNA fragment 369 bp; the segment confirmed by sequencing was the purpose of amplification products. Results Used to establish the method of FTA filter membrane combination of PCR detection Shigella in the meat. The detection limit of the PCR were all 8× 101CFU /ml of meat homogenates of artificial contamination of beef,lamb,pork and chicken,the whole detection process can be finished within 6 hours. Conclusions This method is sensitive and fast,to provide new detection way for prevent Shigella poisoning,control the invasion of bacillary dysentery.
