摘  要:许多研究表明,水母能够耐受低氧环境,但是并不清楚其生理与分子机制.本研究根据海月水母转录组测序结果,获得了海月水母低氧诱导因子(HIF)的部分基因序列,利用实时荧光定量PCR方法检测海月水母HIF基因在低氧条件下(溶解氧约为0.5 mg L-1)的表达变化情况.结果表明,在低氧条件下,海月水母幼体HIF-1α的基因相对表达量经过一定时间的培养后显著高于对照组;以管蛋白基因为内参基因,HIF-1α基因相对表达量在培养3,6,12,18,48和72 h时分别为参照组(饱和溶解氧)的83.2%,102.4%,122.8%,301.3%,140.3%和150.7%.表明低氧环境中海月水母HIF基因表达量在转录水平有极为显著的增加,这为从分子水平揭示水母耐受低氧环境的机制提供了新的视点.Many studies have reported that jellyfish can tolerate low oxygen environments, but the mechanism by which this occurs has not been clarified. In this study, the partial gene of hypoxia inducible factor (HIF-1 a) from Aurelia. sp. 1 was obtained by transcriptome sequencing analysis. The expression of HIF-la in A. sp.1 juveniles was then measured after cultivation in filtered seawater with 0.5 mg L-1 dissolved oxygen. The relative gene expression of HIF-1 ct decreased to 83.2% relative to the ct-tubulin after 3 h when compared with a control group cultured in seawater with saturated dissolved oxygen, then increased to 102.4% after 6 h, 122.8% after 12 h, 301.3% after 18 h, decreased to 140.3% after 48 h, and finally stabilized at 150.7% after 72 h. When the samples were treated for 12 h in hypoxia and then cultivated in seawater with saturated dissolved oxygen for another 12 h, the relative gene expression" of HIF-la decreased to 74.7% relative to the reference group. Overall, these findings indicate that the gene expression of HIF from A. sp. 1 varied obviously under hypoxic conditions.
