摘 要:对近年来视黄素受体介导视黄素抗癌作用机理的研究进展作一综述 ,主要有 :1)视黄素受体 ;2 )视黄素受体与细胞生长 ;3)视黄素受体与孤生受体的相关性 ;4 )视黄素受体对 AP- 1活性的抑制作用 ;5)视黄素受体介导的信号转导途径 .通过研究 ,对于探讨视黄素受体的功能、阐明视黄素的抗癌机理、合成更多的受体选择性视黄素具有重要的现实意义 .Retinoids are a group of natural and synthetic vitamin A analogs that exert profound effects on the growth and differentiation of normal, premalignant and malignant epithelial cells in vivo and in vitro. Retinoids are currently being employed in the clinical treatment of many forms of cancer. The effects of retinoid are mainly mediated by two classes of nuclear receptors: the retinoic acid receptors (RARs) and retinoid X receptors (RXRs). RARs and RXRs function as ligand activated transcription factors that bind to specific retinoic acid responsive sequences on the target genes, and regulate the transcriptional expression of these genes. These receptors, thus, display distinct patterns of expression and have specific functions. In this paper, we summary progress made by ours and others from some aspects as below: 1) retinoid receptors; 2) effect of retinoid receptors on the growth inhibition of cancer cells; 3) relationship between retinoid receptors and orphan receptors; 4) effect of retinoid receptors on the inhibition of AP 1 activity; and 5) signal transduction pathway mediated by retinoid receptors. Our aims for these studies are to evaluate the role of retinoid receptors in mediating anti cancer effect of retinoids in various cancer cells, which may be helpful for exploring the mechanism of retinoids on the trial of malignant tumor, and may put forward a new strategy for synthesis of receptor selective retinoids.
