表面活性剂对GD_3合成酶活性的影响及增溶作用

摘  要:应用不同类型的表面活性剂系统研究其对大鼠肝癌GD_3合成酶(CMP-唾液酸-GM_3唾液酸转移酶,ST2)活性的影响及增溶作用。研究结果表明Triton系列的表面活性剂增溶作用较好,在0.1mol/L KCl存在下Tritoa CF-54和Triton X-100(1:1V/V)两者混溶时,其增溶效果比其单独使用更佳,非离子型表面活性剂NP-40和Triton CF-54具有最大的激活作用。当离子型表面活性剂SDS的胆酸钠浓度在0.1%~0.2%时对ST2略有激活作用,面大于0.4%时有抑制作用。不同的表面活性剂对SF2活性产生不同的影响,提示SF2与脂结合的结构区域,与表面活性剂或脂结合时对ST2活性具有调节作用。The effect and solubilization of CMP-NeuAc-GM3 sialyltransf erase (ST2) by various detergents were studied in rat hepatoma. The results showed that when Triton CF-54 was integrated with Triton X-100 (1:1, W/V), the solubilization of the detergents was much more effective than that of Triton OF-54 or Triton X-100 alone. The maximum extraction rate of the desired ST2 activities was over 95%. The nonionic detergents NP-40 (0.2% W/V} and Triton OF-54 (0.2%, W/ V), compared with other nonionic detergents, produced the greatest enzyme activation, which provided a 12-fold and 11-fold stimulation, respectively. Conversely, ionic detergents SDS (>0.4%), sodium cholate (>0.4%) inhibited the enzyme. Furthermore it was found that potassium chloride enhanced the solubilization of membrane protein, while the specific activity and the recovery rate of total activity of ST2 decreased with the increase of potassium chloride. The various effects of detergents on ST 2 activity Suggest that a lipid binding domain is present in ST 2 and a combinaiton of ST 2 and a detergent or lipid might modulate the activities of ST2.
