摘 要:作者对小鼠肝癌肿瘤浸润性淋巴细胞(Tumor-infiltrating lymphocyte,TIL)的抗瘤活性进行了初步研究。结果表明,新鲜分离的TIL对自身肝癌细胞的杀伤效应很低,经与IL-2共育后显著提高。培养10~30天达高峰,以后开始下降,灭活自身肿瘤细胞的再刺激,可对抗这种下降趋势。TIL对自身肝癌细胞的杀伤后性明显强于LAK细胞(P<0.01),并且表现为较强的肿瘤特异性,即对自身肿瘤细胞的杀伤明显强于对其它肿瘤的杀伤。将TIL与肿瘤细胞混合(10:1)注入小鼠皮下,肿瘤细胞的生长受到明显抑制。上述结果提示,TIL可望成为肿瘤过继免疫治疗中较LAK细胞更有效的活性细胞。Antitumor activities of tumor-infiltrating lymphocytes (TILs) from mouse hepa-toma had been studied. The in vitro results showed that (1) freshly isolated TILs hada poor cytotoxicity against autologous hepatoma cells, an obvious enhancement of theirlytic ability was demonstrated in the presence of IL-2 and maximum lysis was ob-served at 10-30 days in culture; (2) restimulation of long-term TIL culture with in-activated autologous hepatoma cells could restore and maintain the cytotoxicity of TILswhich had been declined 30 days after cultured with IL-2 alone; (3) TILs had a high-er capacity to kill autologous tumor cells than LAK cells. In vivo, TILs could signi-ficantly suppress the subcutaneous growth of hepatoma cells when they were inoculatedwith TIL to tumor cell ratio at 10:1. These observations suggested that TILs might bea good effector in adoptive immunotherapy for malignancies.
