β-类淀粉样蛋白放免分析法及其在老年痴呆症中的初步应用

摘　　要：本文利用 β 类淀粉样蛋白 (β -AP)片断与甲状腺球蛋白经戊二醛偶联为免疫原 ,免疫新西兰兔 ,6个月后获得抗体 ,建立了高灵敏度的 β -AP放射免疫分析法。批内和批间CV分别为 8%和 14%。平均回收率为96 %。 41名正常人血清 β AP浓度为 39± 5 .5 pg/mL。 6 0例老年痴呆症患者 ,血清β AP浓度明显高于对照组 (P<0 .0 1)。本文结果表明 ,测定血清 β -AP浓度 ,可作为一项检查老年痴呆症的指标。Rabbits were immunized with beta-amyloid protein(β-AP)coupled with thyroglobulin(TG).After six months a specific antiserum was obtained. A sensitive RIA was developed for the measurement of β-AP with intra and inter assay CV of 8% and 14% respecively. The recovery rate was 96%. The levels of serum β-AP in 41 healthy adults were 39±5.5pg/mL. The serum β-AP concentrations in 60 patients with AD were higher than those of control group. Serum β-AP level may be an indication for the diagnosis of AD pateints.
