关节Ⅰ号方治疗骨关节炎的实验研究

摘　　要：为阐明补益肝肾中药复方关节Ⅰ号方治疗骨关节炎 (OA)的作用机理 ,参照Colombo方法造成家兔膝OA模型。并在对侧膝关节做假性手术对照。将模型兔分为空白对照 (A)组、关节Ⅰ号方治疗 (B)组、壮骨关节丸对照 (C)组 ,4W后对照观察各组病变关节的超微结构 ,并测定相关生化指标。结果 :关节Ⅰ号方的消炎消肿作用明显优于壮骨关节丸 ,还能显著提高模型兔关节滑膜cAMP、关节软骨氨基多糖及天冬氨酸、谷氨酸、甘氨酸、颉氨酸、蛋氨酸、苯丙氨酸、赖氨酸 7种氨基酸的含量 ,与两对照组相比有统计学差异 (P <0 0 5～ 0 0 1)。提示关节1号方能促进OA早期基质合成代谢和软骨修复。In order to explain the functional mechanism of “Compound Arthritis No.1 Formula”prepared with the herbal drugs to tonify and benefit the liver and kidney in the treatment of osteoarthritis(OA),OA models were made by Colombo method in the rabbit knees,with pseudosurgery done on the opposite knee joints for comparison.The model rabbits were divided into the three groups of the empty control group(A),treatment group(B)of “Arthritis No.1 Formula”and control group of “Bone-Strengthening Arthritis Pills”,to observe and compare the ultrastructure of the affected joints and determine the related biochemical indexes after 4 weeks ih various groups.The results showed that “Arthritis No.1 Formula”was obviously better than “Bone-Strengthening Arthritis Pills”in eliminating inflammation and diminishing swelling and was also able to remarkably enhance cAMP in the articular synovium in the rabbits,amino-polysaccharide in the articular cartilage,and contents of seven amino acids of aspartic acid,glutamic acid,glycine,valine,methionine,phenylalanine and alanine,with a statistic difference in comparison of the two control groups(P<005～0.01).The findings indicate that “Arthritis No.1”can promote the synthetic metabolism of the stromata in the early OA and cartilage repair.
