淋巴滞留性脑水肿的动物模型

摘　　要：目的建立一种简便易行、重复性好、符合人类发病条件的淋巴滞留性脑水肿模型。方法采用阻断大鼠脑淋巴引流的方法 ,建立淋巴滞留性脑水肿的动物模型 ,观察其行为学变化 ,并从术后不同时间取脑标本 ,进行肉眼、普通光镜、透射及扫描电镜观察 ,证实脑水肿的存在。结果所有动物均发生脑水肿 ,表现行为异常 ,脑膜及脑组织肿胀、充血 ,小血管壁外膜及Virchow -Robin间隙增宽 ,内含大量水肿液 ,毛细血管基底膜肥厚 ,神经元固缩、轮廓不清晰或肿胀、坏死 ,胶质细胞增生、肿胀 ,硬脑膜表面凹凸不平、水肿明显。结论阻断脑淋巴引流 ,可以制备淋巴滞留性脑水肿模型 ,其操作简便、成功率高、重复性好。Objectve To establish a simple easy-handle and repeated model of LCE in rats which fits to a condition of diseases in human. Methods The model of LCE was established by occluding lymphatic drainage of the brain in rats. After blocking the animals behavioral alterations were observed, and their brain tissues were studied 1d, 2d, 3d, 5d and 7d after the operation, at the gross level, the light-microscopic level and electron-microscopic level(transmission and scanning) to confirming the existence of cerebroedema. Results Cerebroedema appeared in all animals and they have some behavioral alterations. The meninges tighten, the brain swollen overtly. The adventitia of the brain arteries and the perivascular Virchow-Robin space dilated irregularly. In some neurons both the nucleus and cytoplasm condensed, chromatin gathered near the nuclear membrane. Some other neurons appeared degeneration or necrosis. There was intracelluar edema of the astrocytes.Conclusion LCE animal model which was made by blocking the drainage of cerebral lymphatic is easy. It can be repeated and fits to character of human LCE.
