皮肤鳞状细胞癌和基底细胞癌癌细胞表达CD54抗原的免疫电镜研究

摘  要:为探讨皮肤鳞状细胞癌 (鳞癌 )和基底细胞癌 (基癌 )癌细胞上是否低水平表达 CD54抗原,应用低温聚合、包埋后染色的胶体金免疫电镜技术,对 3例鳞癌和 3例基癌 CD54免疫组化染色阴性的癌细胞进行了研究。在 3例鳞癌和 3例基癌中,各 2例癌细胞表面及突起处观察到 CD54胶体金颗粒的存在;其中 1例鳞癌张力微丝-桥粒复合体处还可见 CD54胶体金颗粒分布。研究发现,虽然鳞癌和基癌癌细胞 CD54免疫组化染色阴性,但应用胶体金免疫电镜可以观察到其癌细胞低水平表达 CD54抗原。该结果可解释皮肤鳞癌和基癌临床上可发生转移,但发生率较低的机理,值得进一步研究。To investigate whether there was a low level expression of CD54 antigen on tumor cells with negative CD54 immunohistochemical staining in squamous cell carcinoma(SCC) and basal cell carcinoma(BCC), tumor cells with negative CD54 immunohistochemical staining in each 3 cases of SCC and BCC were studied by employing low-temperature polymerization and post-embedded staining immunoelectron microscopic(IEM) technique. The CD54 colloid gold particles were labeled on the cell surface or projections of tumor cell membrane in each 2 cases of SCC and BCC; and in one case of SCC, CD54 colloid gold particles were also detected on tonofilament desmosome complex. It was found that: although the tumor cells of SCC and BCC were negative CD54 immunohistochemical staining, but the low level expression of CD54 antigen on the tumor cells could be observed with the use of colloid gold IEM. The results might explain the mechanism why metastasis may occur, but with low incidence in some of skin SCC and BCC.
