摘 要:目的 胶原是骨生成过程中最重要的基质成份 ,本实验探究牵拉成骨过程中胶原生成及演变的机理和规律。方法 运用改良的 Neum an和 L ogan方法对牵拉间隙新骨形成过程中基质胶原总量的变化进行测量 ;运用免疫组织化学分析技术 ,在蛋白质水平对 、 型胶原在不同时间和空间上的出现作一系统的分析研究。结果 胶原在牵拉后不久即出现 ,早期胶原分泌旺盛 ,牵拉完成后 2周即达正常的 6 7%。牵拉间隙早期新骨中有丰富的 型胶原 ,少量的 型胶原 ;而中、晚期发现大量的 型胶原构成骨小梁的主要基质成份 , 型胶原已很少测出。结论牵拉早期间隙中成纤维细胞分泌 型胶原 ,成骨细胞在此基础上分泌沉积 型胶原 ,同时骨小梁不断钙化、改建 , 型胶原逐步降解 ,最终 型胶原代替Objective Collagen is the main matrix during the period of osteogenesis. The aim of the present study was to characterize the changes in collagen types during distraction osteogenesis. Methods Osteotomy of the mandibles followed by gradual distraction was performed on 12 sheep. We analysed sequential biopsies obtained from the center of the distraction gaps during the first 8 weeks of distraction. The amount of collagen of the gap was measured with modification of Neuman and Logan method. Determination of the type of fibrillar collagen was performed by immunohistochemistry which is particularly useful in studies of the distribution of the various types of collagen in bone. Results The whole distraction area was rapidly filled with organic martrix. Collagen appeared in the distraction area in the early phase of distraction osteogenesis and increased rapidly after 2 weeks of distraction complated. The amount of total collagen is 67% of the level in control bone at 2 weeks. Collagen synthesis and organization of the collagen bundles accord to the direction of distraction precede bone formation. There was plenty of type Ⅲ collagen in the early phase, but there was few type Ⅰ collagen. After 8 weeks, the matrix of new bone is composed of type Ⅰ collagen without type Ⅲ collagen. Conclusions Type Ⅲ collagen was detected at the distraction area in the early stage of distraction osteogenesis. Soon afterwards a great quantity of type Ⅰ collagen was secreted and it gradully became main portion of matrix instead of type Ⅲ collagen.
