摘 要:目的 观察神经元和神经胶质细胞标志物 PGP 9.5和 S-1 0 0蛋白在先天性巨结肠 (hirschsprung disease,HD)中的表达。方法 采用 PAP免疫组织化学方法。结果 (1 )在对照组结肠壁神经丛中可见染色深浅不一的PGP9.5免疫反应性神经节细胞 ,神经纤维均匀分布在肠壁各层 ,并与肌纤维相平行。神经节细胞胞体对 S-1 0 0蛋白则表现为细胞状“空白区”。(2 ) HD结肠壁分化异常 ,PGP 9.5和 S-1 0 0蛋白免疫反应性神经纤维明显增生 ,分布紊乱 ,未见有 PGP9.5阳性神经节细胞。在增生的 S-1 0 0蛋白阳性神经纤维中偶见有细胞状的“空白区”。结论 神经丛中 PGP 9.5阳性反应的细胞团块和 S-1 0 0蛋白染色的神经丛中细胞状“空白区”,可特征性地提示神经节细胞的存在。实验证实 ,结肠壁神经发育异常是 HD的主要病理生理变化。Objective To observe the expression of PGP 9.5 and S-100 protein in the colon of the patients with Hirschsprung's disease. Methods Immunohistochemical method was used in the experiment. Results (1) In control group, neurons with different staining were observed in the muscle layer and positive nerve fibers paralleled with the muscle fibers. The neurons were stained negatively to S-100 protein. (2) In the HD group, both PGP 9.5 and S-100 protein positive fibers were proliferated obviously in number and wave-like in their appearance. Although no PGP 9.5 positive neurons were found in the plexus,the cell-like blank areas were seldomly observed in the hyperplasia S-100 protein positive neuroplexus. Conclusions The PGP 9.5 immunoreactive neurons and S-100 protein immunoreactive cell-like blank area demonstrate the lying of neurons in myenteric plexus.However, this experiment, has proved the pathophysiological changing of HD.
