体外培养大鼠星形细胞的缺糖缺氧性损伤及药物的保护  被引量:4

摘  要:本实验用大鼠星形细胞体外培养模型,观察了细胞在不同条件下乳酸脱氨酶(LDH)的漏出。发现当去掉葡萄糖后,细胞缺氧5和7h后LDH漏出明显增加,5b为59.7±25.3U/mg蛋白(对照组24.7±16.3,P<0.05),7h为68.3±89U/mg蛋白(对照组39.9±212,P<0.01)。用3-氧-甲基-[1- ̄3H]-D-葡萄糖摄取法测量细胞体积,结果显示缺糖缺氧5h后细胞明显肿胀,从对照组的4.1±1.2增加到8.1±3.2μl/mg蛋白(P<0.01)。丹参有效成分764-3在0.5至50μmol/L时能减少缺糖缺氧细胞LDH漏出;在50μmol/L时能减小缺糖缺氧细胞的体积。谷氨酸受体拮抗剂DNQX(6,7-dinitroquinoxaline-2,3dione)50μmol/L能减轻星形细胞肿胀和LDH漏出。The effects of severe O_2 and glucose deprivation were studied in a subculture of astrocytesprepared from neonatal rat cerebral cortices.After two weeks of incubation in vitro,culturedastrocytes were further incubated with Earle's solution and subjected to hypoxic condition at95% N_2+5% CO_2 for3,,7h.When glucose was taken out from the culture medium,significantcell damage occurted at 5 and 7h of hypoxia,as evidenced by a time,dependent leakage of lactatedehydrogenase(LDH), being 59.7+ 25.3U/mg protein at 5h (24.7± 16.3 in control,p<0.05)and 68.3 ± 8.9 U/mg protein at 7h(39.9 ± 21.2 in control,P<0.01),The cell volume ofastrocytes was measured with 3-O-methyl-[1-3H]-D-glucose uptake. The volume of astrocytesincubated for 5h without glucose and oxygen increased remarkably,from 4.1±1.2μl/mg proteinin control group to 8.1 ± 3.2 μl/mg protein(P<0.01).Under this condition 764-3.an activecomponent of Chinese medicinal herb Salvia mIjliorrhiza Bge,(50μmol/L) or 6,7-dinitroquinoxalin-2,3-dione(DNQX),an antagonist to AMPA/KA subtype of glutamatereceptors,(50μmol/L)significantly decreased cell swelling and the leakage of LDH.
