摘 要:目的 观察土牛膝液对宫颈癌热敏感性的影响。方法 用细胞计数集落形成率 ,了解癌细胞的热敏感性 ,用光镜和流式细胞术观察凋亡细胞 ,免疫组化法观察细胞 HSP70蛋白合成情况。结果 经土牛膝预先处理的细胞增高了对热处理的敏感性 ,癌细胞发生了凋亡 ,细胞膜 HSP70蛋白的表达率下降 ,这些效应均与土牛膝的处理剂量相关。结论 土牛膝液能增强对宫颈癌热敏感性 ,增强这种热敏感性的可能机理是通过诱导细胞凋亡和抑制 HSP70蛋白的合成。Objective To investigate the thermosensitivity effect of achyranthes aspera on cervical cancer cells.Methods Cervical cancer line Hela cell was used to study its sensitivity to heat(42℃,lhr)treatment.Heat\|induced changes in cell viability was assessed by colony formation assay,apoptosis by cell morphology and flow cytometry,the sythethtic HSP70 protein by the expression of membrane protein HSP70 of cells.Results With achyranthes aspera on the Hela cells the thermosensitivity and the percentage of apoptotic cells were significantly increased, the expressin of membrane protein HSP70 of cells was decreased. These effects were dependend on the dose achyranthes aspera.Conclusion The thermosensitivity of Hela cells can be increased by achyranthes aspera.Apoptosis and HSP70 protein may be involved in the increased in thermosensitivity.
