交链孢酚单甲醚对鼠肝脂质过氧化及谷胱甘肽过氧化物酶的影响

摘  要:用食管癌高发区林县的粮食中分离出的互隔交链孢霉的代谢物之一——交链孢酚单甲醚(AME)注入小鼠体内,观察对鼠肝中脂质过氧化及谷胱甘肽过氧化物酶活性的影响。试验结果显示:AME可以导致丙二醇含量增加(P<0.01),谷胱甘肽过氧化物酶活性降低(P<0.01)。丙二醛含量的增高与AME剂量呈显著正相关(r=0.957,P<0.05),酶活性与丙二醛水平呈明显负相关(r=-0.973,P<0.05)。结果表明:AME可以引发脂质过氧化反应,谷胱甘肽过氧化物酶活性下降,本实验结果为该霉菌极可能是引起食管癌病因之一学说提供的积极佐证。Alternaiol Monomethyl ether(AME) produced by Alternaria alternate was isolatedfrom the grains in areas of high incidence of esophageal cancer in Linxian Countyand injected into mice. The effect of AME, a suspected carcinoger in human being,on the lipoperoxidation and GSHPX activity in mouse liver was studied.The result showed that AME resulted in a reduction of GSHPX activity (p<0.01). and an increase of the MDA levels (p<0.01) There existed a positive correla-tion between the MDA levels and the AME dosages,(r=0.957, p<0.05), GSHPX acti-vity showed significant regative correlation with MDA levels(r=-0.973, p<0. 05)Itwas demonstrated that the lipoperxidation could be initiated with AME. The results sho-wed that AME might be one of the factors in the carcinogenesis of esophageal cancer in thearea.
